Short description:

Cryo-microscopy is becoming an increasingly popular method of obtaining information about organic macromolecules such as proteins, RNA, and DNA, as well as, larger objects such as viruses, liposomes, bacteria, and tissue fragments. Due to the growing demand for studies on objects of various sizes, often at the limit of the applicability of the technique, there is a need to investigate and systematize the impact of imaging parameters on the data quality and the reconstruction resolution that can be obtained. These are parameters such as, among others, magnification, defocus, dose and data collection scheme. The research covered by the thesis will focus on issues in the field of image formation, the information limit in transmission electron microscopy and the influence of the mentioned parameters on previously mentioned mechanisms.

Requirements to the candidate: 

  • knowledge of the topics related to the interaction of electrons  with matter and electron imaging techniques
  • English language skills enabling the presentation of scientific results in written and oral form
  • experience with research equipment, especially with cryo-EM
  • master’s degree in physics, chemistry, material sciences, or a related field
  • experience in cryo-EM sample preparation and microscope handling
  • knowledge of the topics related to experimental data analysis and correlation between the results and measurement parameters
