G protein coupled receptors (GPCRs) are a large family of membrane-bound receptors that mediate the majority of physiologic responses to hormones and neurotransmitters, as well as the senses of sight, smell, and taste. GPCRs are the largest group of pharmaceutical targets for a broad spectrum of diseases. The goal of my research over the past 32 years has been to characterize the dynamic process of GPCR activation by applying a spectrum of biophysical and structural approaches. I will discuss what these studies have taught us about GPCR signaling and how structural approaches can advance drug discovery.
